True QPI: Metrology of Quantitative Phase Imaging Systems for Biomedical Applications

The proposed Experience Centre is a unique training that allows participants to learn how to perform metrological assessment of quantitative phase imaging (QPI) systems based on bio-inspired phantoms manufactured with two photon polymerization.

The training will be supplemented with a short 3-hour lecture on the theoretical basics of the techniques used. In this way a complete package on the QPI systems metrology will be delivered during the training. The course is aimed at 9 participants – this constitutes 3 groups of 3 participants – which is a maximum number we found to be able to benefit from the hands-on training style. The training level is suitable for people with general knowledge in photonics and especially microscopy. The participants will have access to two QPI laboratories and a microfabrication facility.

Learning outcomes

After completing the module, participants will be able to:

  1. Explain the operating principles of major QPI modalities (digital holography, ptychography, tomographic phase imaging),
  2. Describe the metrological challenges of QPI systems, including calibration needs, measurement uncertainty, and the role of standardized phantoms,
  3. Understand the relationship between phase, refractive index, and biological structure, enabling meaningful interpretation of QPI data,
  4. Operate multiple QPI instruments (2D and 3D) and configure them for biological sample imaging.
  5. Perform full metrological validation workflows, including calibration, alignment, and performance assessment of QPI systems.
  6. Acquire, reconstruct, and analyze QPI datasets, including phase retrieval, tomographic reconstruction, and extraction of quantitative metrics.
  7. Evaluate the performance and limitations of different QPI systems using quantitative criteria and metrological best practices.
  8. Identify sources of measurement error and propose strategies to improve accuracy, repeatability, and reliability.
  9. Select the most appropriate QPI modality for a given biomedical or industrial task based on sample type, resolution needs, and measurement goals.
